Madekwe, a mixed race British-Nigerian child was born into a family in South London. Her father and mother have both been English. She went to The Brit School when she was fourteen years old. It was the Royal Academy of Dramatic Art is where she earned the BA in acting. While at school she appeared in several stage productions, including King Henry V's princess Katherine as well as Wuthering Heights.

Madekwe's US debut was in 2009, with the drama The Beautiful Life. The show aired just two episodes before being removed from The CW. Ashley Davenport, a character on ABC's Revenge drama series was cast to play Ashley Davenport. Madekwe was a regular on the ABC drama series Revenge for two seasons. In May 2013 that she'd be leaving the series following an appearance in the initial episode of season three. In 2014 and 2016, she starred as Tituba the television series Salem. Madekwe got married to her long-time lover, Iddo. They have worked together in the show Secret Diary of a Call Girl and also had a role alongside her in the television series Salem.

Ashley Leggat, an actress that was nominated two times for a Gemini Award, is a member of the large Hamilton family. Her parents Roy and Patty and the four brothers of her, Brett, Todd, and Bram, were all present.
